HCY™ Pfu DNA Polymerase 

Pfu DNA Polymerase, abbreviated as Pfuase, is one of the most commonly used high-fidelity DNA polymerases.
HCY™ Pfu DNA Polymerase is a highly heat-stable DNA polymerase derived from the thermophilic bacterium Pyrococcus furiosus, which has been modified by mutation and recombination to have higher thermal stability and amplification efficiency than the wild-type Pfu DNA polymerase.The molecular weight of the Pfu enzyme is 90 kDa.The Pfu enzyme possesses 5‘-3’ DNA polymerase activity and 5‘-3’ exonuclease activity. The molecular weight of Pfu is 90 kDa, and Pfu has 5‘-3’ DNA polymerase activity and 5‘-3’ exonuclease activity, and the PCR product has smooth ends, which can be directly used for flat-end cloning.
Due to the 3‘ to 5’ exonuclease activity of Pfu, the chances of error during PCR amplification are greatly reduced and the fidelity is 10 times higher than that of Taq DNA polymerase.The error rate of Pfu is not only much lower than that of Taq, but also lower than that of some other high fidelity DNA polymerases such as Vent, DeepVent, Pwo, Tli, etc. Therefore, Pfu is often preferred as a cost-effective high-fidelity DNA polymerase.
HCY™ Pfu DNA polymerase has high amplification efficiency and fast amplification speed (4 kb/min, 8 times faster than ordinary Pfu enzyme), which overcomes the defects of ordinary Pfu enzyme’s low amplification efficiency, low yield and slow amplification speed, and greatly shortens the reaction time.

HCY™ Pfu DNA Polymerase is a Recombinant Pfu DNA Polymerase, mutated and recombinant, purified by E. coli expression.
Uses

High-fidelity PCR, point mutation, double-flat-end PCR cloning, etc.
The DNA fragment amplified by Pfu enzyme is double flat-end, which can be used for double flat-end cloning, but cannot be used for conventional T vector cloning.
dUTP and dITP or primers containing dUTP and dITP cannot be used in Pfu enzyme-mediated PCR amplification reactions.
Definition of activation

The activity of 10 nmol of whole nucleotides ingested as acid insoluble material was defined as 1 activity unit (U) at 74°C for 30 min using activated salmon sperm DNA as template/primer.
