- Preparation Work
- Ultra – pure water or deionized water needs to be prepared, because impurities in water may affect the quality of the culture medium. The containers need to be strictly cleaned and sterilized to prevent contamination by impurities and microorganisms.
- Dissolution Process
- According to the ratio of powder to water specified in the product manual, slowly add the cell culture medium powder to the water. Generally, add it while stirring to ensure that the powder is evenly dispersed in the water and prevent lumps from forming. Stirring is usually carried out with a magnetic stirrer or a glass rod, and the stirring speed is moderate to avoid generating too many bubbles.
- Adding Other Components
- According to the specific requirements of cell culture, add other components such as serum, antibiotics, and growth factors to the dissolved culture medium. For example, for most mammalian cell cultures, fetal bovine serum (FBS) is usually added, and the addition amount is generally between 5% and 20%, to provide additional nutrients and growth factors required for cell growth.
- pH Adjustment and Filtration Sterilization
- Use pH test paper or a pH meter to detect the pH value of the culture medium and adjust it with acid (such as hydrochloric acid) or alkali (such as sodium hydroxide) as needed to make it reach the pH range suitable for cell growth (usually between 7.2 and 7.4). Finally, filter the prepared culture medium through a sterile filter membrane of 0.22 μm to remove possible microorganisms and ensure the sterility of the culture medium before it can be used for cell culture.
