Leibovitz L-15 medium supports the growth of HEP-2 monkey kidney cells as well as primary tissue blocks of embryonic and adult tissues.L-15 is buffered with phosphate and free amino acids rather than sodium bicarbonate. This medium is suitable for supporting cell growth in non-CO2 equilibrium environments.L-15 contains no proteins, lipids, or growth factors. Therefore, F-10 requires additives, typically 10% fetal bovine serum (FBS).

This Leibovitz L-15 Dry Powdered Media requires pH adjustment and filtration during preparation. The detailed preparation protocol is as follows:
Add 950 mL of distilled water to a mixing vessel as close to the final volume as possible.
Add the dry powdered medium to room temperature (15°C to 30°C) water and stir gently. Do not heat the water.
Rinse the inside of the package to remove any residual powder.
Adjust pH to 7.6 by slowly adding 1 N NaOH or 1 N HCl and stirring.
Adjust final volume using distilled water.
Immediately dispose of the medium into a sterile container through a 0.2 μm filter membrane using a positive pressure filtration system.
